Some Electrical Properties of Single Renal Tubule Cells
نویسنده
چکیده
At a considerable rate of metabolic expenditure, the renal tubules transfer large amounts of ions across their epithelial cell layer. Recently, a better characterization of the nature of individual ionic transport processes at the single nephron level has become possible through an assessment of some electrical characteristics of tubule cells. The response of either transmembrane or transepithelial potential differences to changes in the ionic environment, as well as measurements of the current-voltage relationship across various tubular structures, have served to clarify certain ionic conductance characteristics. In addition, insight into the functional organization of adjacent tubule cells is provided by recent inquiries into the nature of the current distribution along renal tubular structures.

Measurements of Electrical Potentials and Ion Fluxes
Experimental technics are described which permit the measurements of stable electrical potential differences across the cell membrane of single renal tubule cells and across single tubules. The inside of the tubule lumen is normally found to be electrically negative to the outside, while the cell interior is found to be negative to both the peritubular fluid and the tubule lumen. Such electrica...
متن کاملMeasurements of Electrical Potential Differences on Single Nephrons of the Perfused Necturus Kidney
Stable electrical potential differences can be measured by means of conventional glass microelectrodes across the cell membrane of renal tubule cells and across the epithelial wall of single tubules in the doubly perfused kidney of Necturus. These measurements have been carried out with amphibian Ringer's solution, and with solutions of altered ionic composition. The proximal tubule cell has be...
متن کاملMolecular diversity and regulation of renal potassium channels.
K(+) channels are widely distributed in both plant and animal cells where they serve many distinct functions. K(+) channels set the membrane potential, generate electrical signals in excitable cells, and regulate cell volume and cell movement. In renal tubule epithelial cells, K(+) channels are not only involved in basic functions such as the generation of the cell-negative potential and the co...
متن کاملConductive properties of the proximal tubule in Necturus kidney
The electrical properties of the proximal tubule of the in vivo Necturus kidney were investigated by injecting current (as rectangular waves) into the lumen or into the epithelium of single tubules and by studying the resulting changes of transepithelial (VL) and/or cell membrane potential (VC) at various distances from the source. In some experiments paired measurements of VL and VC were perfo...
متن کاملMorphological and functional characterization of a conditionally immortalized collecting tubule cell line.
A conditionally immortalized collecting tubule cell line, mCT1, was derived from the H-2Kb-ts A58 transgenic mouse (ImmortoMouse), which harbors a temperature-sensitive mutant of the SV40 large T antigen oncogene. Cells maintained under permissive conditions [33°C with interferon-γ (IFN-γ)] form epithelial monolayers, express large T antigen, and proliferate (>50 passages). The cells retain pro...
